{"created":"2023-06-19T09:22:24.742125+00:00","id":1648,"links":{},"metadata":{"_buckets":{"deposit":"1c1c7acd-6939-40a1-918b-9271b67aff3e"},"_deposit":{"created_by":4,"id":"1648","owners":[4],"pid":{"revision_id":0,"type":"depid","value":"1648"},"status":"published"},"_oai":{"id":"oai:auhw.repo.nii.ac.jp:00001648","sets":["52:242"]},"author_link":["8993","8992","8998","9005","8996","9003","9004","8994","9000","8995","8999","9001","8997","9002"],"item_10002_alternative_title_1":{"attribute_name":"その他(別言語等)のタイトル","attribute_value_mlt":[{"subitem_alternative_title":"The effect of onion and Ginkgo biloba extract for oxidative stress and cardiac dysfunction on 3'-azido-3'-deoxythymidine (AZT) administered rats"}]},"item_10002_biblio_info_7":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2005-03","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"2","bibliographicPageEnd":"117","bibliographicPageStart":"109","bibliographicVolumeNumber":"6","bibliographic_titles":[{"bibliographic_title":"青森県立保健大学雑誌"},{"bibliographic_title":"Journal of Aomori University of Health and Welfare","bibliographic_titleLang":"en"}]}]},"item_10002_description_19":{"attribute_name":"フォーマット","attribute_value_mlt":[{"subitem_description":"application/pdf","subitem_description_type":"Other"}]},"item_10002_description_5":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"3'-azido-3'-deoxythymidine (AZT) は, エイズの治療薬として用いられているが, 心臓等に酸化ストレスを引き起こして心機能を低下させる。そこで, AZTの副作用である心機能の低下を抗酸化食品であるタマネギやイチョウ葉エキスを摂取することで抑制することが可能かどうかを調べた。酸化ストレスを引き起こしやすくするために Fischer-F344系雄性ラットに脂肪分12%の高脂肪食 (High fat diet : HF) を2ヶ月齢から与え12ヶ月齢まで飼育した。その後HFに、0.5%タマネギ凍結乾燥粉末 (HF+Onion : HF+O) または0.2%イチョウ葉エキス (HF+Ginkgo biloba extract : HF+G) を含む各飼料を摂取させた。そして17.5ヶ月齢から1日1回2週にわたってAZTを投与 (40mg/kg) した。最終投与3日後に心電図検査を行い、その後屠殺、解剖して心臓の過酸化脂質 (Lipid Peroxide : LPO) 及びグルタチオン濃度を調べた。同時に心機能と関連が深い血中脂質成分も調べた。その結果、(1) 心電図では各群ともAZT投与群は非投与群と比べて心機能の低下が見られたが、各食餌間に有意な差は見られなかった。(2) 心臓のLPOやグルタチオンの濃度はHF群ではAZT投与をしても変化は見られなかった。しかしながら、AZTを投与したHF+O群では、非投与HF+O群と比べてグルタチオン濃度が有意に増加し、酸化ストレスを低下させていることが示唆された。(3) AZTを投与したHF群は、非投与HF群より血中総コレステロール (T-Ch), HDL-コレステロール (HDL-Ch), LDL-コレステロール (LDL-Ch) および過酸化脂質 (LPO) 濃度が増加した。しかしながらAZTを投与したHF+G群はAZTを投与したHF群と比べて、T-Ch, LDL-Chおよび動脈硬化指数が改善していた。以上の結果から, AZT投与による心臓の酸化ストレスに対してはタマネギが, 血中脂質成分の変化に対してはイチョウ葉エキスがそれぞれ有効であることが示唆された。\nWhile 3'-azido-3'-deoxythymidine (AZT) is being used as a therapeutic agent for AIDS, it induces oxidative stress in the heart and reduces cardiac function. The present study thus examined whether reduced cardiac function associated with AZT administration can be ameliorated by ingesting antioxidant foods such as onion and Ginkgo biloba extract. In order to facilitate induction of oxidative stress, male Fischer-F344 rats were given a high fat (12%) diet (HF) from 2- to 12-months-old. A HF including either 0.5% lyophilized onion powder (HF+O) or 0.2% Ginkgo biloba extract (HF+G) was then given to the rats. Furthermore, from 17.5-months-old, AZT 40 mg/kg/day was administered for 2 weeks. Electrocardiography (ECG) was performed 3 days after the final administration. The rats were euthanized and levels of cardiac lipid peroxide (LPO) and glutathione were measured. Serum lipid components, which are closely related to cardiac function, were measured at the same time. The results were as follows : 1) ECG showed no significant difference between diet groups, although cardiac function tended to be lower for rats with AZT than for rats without. 2) Levels of cardiac LPO and glutathione for the HF group were unchanged by AZT administration. However, glutathione levels were significantly higher for the HF+O group with AZT than for the HF+O group without AZT, suggesting that onion suppresses oxidative stress. 3) Levels of plasma total cholesterol (T-Ch), HDL-cholesterol (HDL-Ch), LDL-cholesterol (LDL-Ch) and lipid peroxide (LPO) were significantly higher for the HF group with AZT than for the HF group without AZT. However, compared to the HF group with AZT, T-Ch, LDL-Ch and Atherogenic Index (AI) were lower for the HF+G group with AZT. These findings suggest that onion is effective against cardiac oxidative stress, and Ginkgo biloba extract is effective in preventing changes to serum lipid components associated with
